Regulation of human salt-sensitivite hypertension by myeloid cell renin-angiotensin-aldosterone system

چکیده
Introduction: Salt sensitivity of blood pressure is a phenomenon in which changes according to dietary sodium intake. Our previous studies found that high salt activates antigen presenting cells, resulting the development hypertension. The mechanisms by salt-induced immune cell activation regulated are unknown. In current study, we investigated effects on renin-angiotensin-aldosterone system (RAAS) gene expression myeloid cells and their impact sensitive hypertension humans. Methods: We performed both bulk single-cell sequencing analysis with vitro vivo treatment humans using rigorous salt-loading/depletion protocol phenotype salt-sensitivity pressure. also measured plasma renin aldosterone radioimmunoassay. Results: while exposure downregulated renin, binding protein receptor, there were no significant genes renin-angiotensin response loading depletion . Plasma individuals tended be lower blunted loading/depletion challenge as previously reported. Discussion: These findings suggest unlike systemic RAAS, acute intake do not regulate RAAS cells.

Molecular basis of salt sensitivity in human hypertension. Evaluation of renin-angiotensin-aldosterone system gene polymorphisms.
We analyzed the association between salt sensitivity in essential hypertension and 8 genetic polymorphisms in 6 genes of the renin-angiotensin aldosterone system. Seventy-one patients with essential hypertension were classified as salt sensitive or salt resistant by means of the 24-hour ambulatory blood pressure (BP) change to high salt intake. The polymorphisms evaluated correspond to the foll...

To define the role of the renin-angiotensin-aldosterone system in a novel salt-sensitive model, neonatal Wistar rats were given capsaicin (50 mg/kg sc) on the first and second days of life. After weaning, male rats were divided into the following six groups and treated for 3 wk with: control + normal sodium diet (CON-NS), CON + high-sodium diet (CON-HS), CON + HS + spironolactone (50 mg x kg(-1...

We investigated the effect of cyclooxygenase (COX) activity on the regulation of the renin-angiotensin-aldosterone system by salt intake. Therefore, Sprague-Dawley rats were subjected to different salt diets [0.02, 0.6, and 8% NaCl (wt/wt)] and treated with the selective COX-2 inhibitor rofecoxib (10 mg x kg body wt(-1) x day(-1)) or with ketorolac at a dose selective for COX-1 inhibition (2 mg...

Chronic heart failure (CHF) is associated with endothelial dysfunction. Activation of the renin-angiotensin-aldosterone system (RAAS) is believed to be important in the deterioration of endothelial dysfunction in CHF through stimulation of oxidative stress. Whereas angiotensin-converting enzyme inhibitors (ACE-I) improve endothelial function in CHF, the effects of angiotensin II AT 1-receptor b...
